Chief Justice Roberts a victim of credit card fraud

WASHINGTON (Reuters) - Chief Justice John Roberts, who spent the last two days presiding over high-profile oral arguments on gay marriage, has been a victim of credit card fraud, a Supreme Court spokeswoman said on Thursday.

Roberts' credit card problem was first reported by the Washington Post in a story that said the chief justice was heard talking about it in a suburban Maryland Starbucks on Tuesday morning. That was the day the court weighed the validity of Proposition 8, a California ban on gay marriage.

Roberts was again in the spotlight on Wednesday when the court weighed the federal Defense of Marriage Act, which restricts the definition of marriage to opposite-sex couples for the purposes of federal benefits.

Kathy Arberg, the court spokeswoman, said Roberts had no comment on the incident.

Details of gene pathways suggest fine-tuning drugs for child brain tumors

Pediatric researchers, investigating the biology of brain tumors in children, are finding that crucial differences in how the same gene is mutated may call for different treatments. A new study offers glimpses into how scientists will be using the ongoing flood of gene-sequencing data to customize treatments based on very specific mutations in a child's tumor.

"By better understanding the basic biology of these tumors, such as how particular mutations in the same gene may respond differently to targeted drugs, we are moving closer to personalized medicine for children with cancer," said the study's first author, Angela J. Sievert, M.D., M.P.H., an oncologist in the Cancer Center at The Children's Hospital of Philadelphia.

Sievert, working with co-first author Shih-Shan Lang, M.D., in the translational laboratory of neurosurgeon Phillip Storm, M.D., and Adam Resnick, Ph.D., published a study ahead of print today in the Proceedings of the National Academy of Sciences.

The study, performed in cell cultures and animals, focused on a type of astrocytoma, the most common type of brain tumor in children. When surgeons can fully remove an astrocytoma (also called a low-grade glioma), a child can be cured. However, many astrocytomas are too widespread or in too delicate a site to be safely removed. Others may recur. So pediatric oncologists have been seeking better options---ideally, a drug that can selectively and definitively kill the tumor with low toxicity to healthy tissue.

The current study focuses on mutations in the BRAF gene, one of the most commonly mutated genes in human cancers. Because the same gene is also mutated in certain adult cancers, such as melanoma, the pediatric researchers were able to make use of recently developed drugs, BRAF inhibitors, which were already being tested with some success against melanoma in adults.

The current study provides another example of the complexity of cancer: in the same gene, different mutations behave differently. Sievert and her colleagues at Children's Hospital were among several research groups who reported almost simultaneously in 2008 and 2009 that mutations in the BRAF gene were highly prevalent in astrocytomas in children. "These were landmark discoveries, because they suggested that if we could block the action of that mutation, we could develop a new, more effective treatment for these tumors," said Sievert.

However, follow-up studies in animal models were initially disappointing. BRAF inhibitors that were effective in BRAF-driven adult melanomas made brain tumors worse?via an effect called paradoxical activation.

Further investigation revealed how tumor behavior depended on which type of BRAF mutation was involved. The first-generation drug that was effective in adult melanoma acted against point mutations in BRAF called V600E alterations. However, in most astrocytomas the mutation in the BRAF gene was different; it produced a fusion gene, designated KIAA1549-BRAF. When used against the fusion gene, the first-generation drug activated a cancer-driving biological pathway, the MAPK signaling cascade, and accelerated tumor growth.

By examining the molecular mechanisms behind drug resistance and working with the pharmaceutical industry, the current study's investigators identified a new, experimental second-generation BRAF inhibitor that disrupted the cancer-promoting signals from the fusion gene, and did not cause the paradoxical activation in the cell cultures and animal models.

This preclinical work result lays a foundation for multicenter clinical trials to test the mutation-specific targeting of tumors by this class of drugs in children with astrocytomas, said Sievert. As this effort progresses, it will benefit from CHOP's commitment to resources and collaborations that support data-intense research efforts.

The direction of brain tumor research over the past several years reflects some of those data-driven advances, says Adam C. Resnick, Ph.D., the senior author of the current paper and principal investigator of the astrocytoma research team in the Division of Neurosurgery at Children's Hospital. "For years, astrocytomas have been lumped together based on similar appearance to pathologists studying their structure, cell shape and other factors," said Resnick. "But our current discoveries show that the genetic and molecular structure of tumors provides more specific information in guiding oncologists toward customized treatments."

Earlier this year, Children's Hospital announced its collaboration with the gene-sequencing organization BGI-Shenzhen in performing next-generation sequencing of pediatric brain tumors at the Joint Genome Center, BGI@CHOP. The center's sophisticated, high-throughput sequencing technology will greatly speed the discovery of specific gene alterations involved in childhood brain cancers.

This genomic discovery program dovetails with the work of the Childhood Brain Tumor Tissue Consortium, a multi-institutional collaboration recently launched by CHOP, with support from the Children's Brain Tissue Foundation. Because even large research centers may not hold enough tumor tissue specimens to power certain research, the consortium pools samples from a group of institutions, providing an important scientific resource for cooperative studies.

"The better we understand the mutational landscape of tumors, the closer we'll be to defining therapies tailored to a patient's specific subtype of cancer," added Resnick.

    States answer help wanted ad to be drone test site

    LOS ANGELES (AP) ? It's the land where Chuck Yeager broke the sound barrier, where the space shuttle fleet rolled off the assembly line and where the first private manned rocketship climbed to space.

    Capitalizing on Southern California's aerospace fortunes, two rival groups want to add another laurel: drone test range.

    They face crowded competition. In search of an economic boost, more than half the country is looking toward the sky ? expected to be buzzing in the near future with pilotless aircraft.

    Before that can become reality, the Federal Aviation Administration last month put out a call to test fly drones at half a dozen to-be-determined sites before they can share the same space as commercial jetliners, small aircraft and helicopters.

    Fifty teams from 37 states answered, vying to win bragging rights as a hub for unmanned aerial vehicles.

    The military has long flown drones overseas to support troops, spy on enemies and fire missiles. There's a recent clamor to fly them domestically to track the health of crops, fight wildfires in remote terrain, conduct search and rescue after a disaster and perform other chores considered too "dirty, dull or dangerous" for pilots. The expanding use for drones comes amid concerns of a "Big Brother" society.

    The untapped civilian market ? estimated to be worth billions ? has created a face-off, with states perfecting their pitch ? ample restricted airspace, industry connections, academic partners ? not unlike what you might read in a tourism brochure.

    "It's the chance to get in on the ground floor of what may be the next big business," said Peter Singer, a robotics expert at the Brookings Institution think tank in Washington. "The states competing hope it might make them the robotics equivalent of Detroit for automobiles in the 20th century or Silicon Valley for computers."

    Winners will play key roles in helping the government seamlessly transition drones, which are controlled remotely by joystick, into the civilian airspace without crashing into other planes or injuring bystanders.

    Supporters of a Southern California test site point to an existing drone presence. General Atomics Aeronautical Systems Inc., based in the San Diego suburbs, makes the Predator that has circled over Iraq and Afghanistan. Just outside of downtown Los Angeles, AeroVironment introduced the world's first hummingbird spy plane and is developing other tiny drones inspired by biology.

    "From start to finish, you can do your UAV work here," said John Rose of the American Institute of Aeronautics and Astronautics, which co-sponsored a three-day drone conference this week in the Los Angeles area focused on civilian uses.

    There are two competing California bids from airport agencies in Ventura County northwest of Los Angeles and Kern County in the Mojave Desert.

    "If we are successful, it would be an economic stimulus for the region moving forward," said Bill Buratto of the Ventura County Economic Development Association, which is working with county airport officials on a plan to have drones fly from Point Mugu, the site of numerous Navy training exercises.

    Their in-state competitor envisions test flights out of the high desert skies about 150 miles north of Los Angeles and touts its remoteness and access to military and civilian facilities currently doing drone research.

    "You kind of want to be in the middle of nowhere. You don't want to risk being close to a populated area," said Eileen Shibley, who leads the effort for the Indian Wells Valley Airport District.

    Other states have taken a different tact, putting on a united front or partnering with a neighboring state to pool resources.

    Ohio ? the home state of Apollo 11 astronaut Neil Armstrong, Mercury astronaut John Glenn and the Wright brothers ? teamed with Indiana to increase both states' odds. Like California, there is budding drone activity in Ohio, most notably the Air Force's sensor research at Wright-Patterson Air Force Base.

    Joseph Zeis of the Dayton Development Coalition doesn't see this as a competition.

    "When the test site selection is over, we're all collaborating on a single goal" to safely merge drones into the national airspace, said Zeis, who's spearheading the Ohio-Indiana venture.

    The FAA is expected to choose the six drone test sites by year's end.

    The specter of thousands of unmanned eyes swarming the sky in the coming years has unnerved privacy advocates, who fear ordinary Americans would be overzealously monitored by law enforcement, considered one of the top users of the technology in the future. As part of the selection process, test site hopefuls must publish a privacy policy and follow existing privacy laws.

    The Association for Unmanned Vehicle Systems International does not have a favorite. But the voice for the domestic drone industry acknowledged that states hosting test sites would benefit economically.

    In a report published earlier this month, the group said states with an already solid aerospace industry are predicted to gain drone business. But other factors, including location of test sites, will also drive job creation.

    That's why California needs to act fast, said state assemblyman Jeff Gorell, who has been pushing for a test site in his district.

    "This is a great opportunity for California," he said. "We might be able to recapture some of the golden era of aerospace."

    Leading experts disagree on evidence behind prostate cancer screening recommendations

    Mar. 25, 2013 ? Do the results of recent randomized trials justify the recent U.S. recommendation against yearly measurement of prostate-specific antigen (PSA) as a screening test for prostate cancer? That's the topic of debate in a special "point/counterpoint" section in the April issue of Medical Care.

    The recommendation against routine PSA measurement relies too heavily on randomized trial data, according to an article by Ruth Etzioni, PhD, of Fred Hutchinson Cancer Research Center, Seattle, and colleagues. They argue that modeling studies provide a truer picture of the long-term benefits of PSA screening. But Dr Joy Melnikow of University of California, Davis, and colleagues disagree, asserting that randomized trials provide a sufficient level of certainty to recommend against PSA screening.

    Point: Short-Term Trials Don't Reflect Long-Term Risk Last year, the U.S. Preventive Services Task Force recommended against routine PSA measurement to screen for prostate cancer. The recommendation was mainly based on two recent studies -- one conducted in Europe and one in the United States -- in which men were randomly assigned to annual PSA screening or no screening. Both studies concluded that annual screening did not reduce the risk of death from prostate cancer.

    But randomized trials have important limitations as a basis for screening policies, according to Dr Etzioni and colleagues. They note that screening trials generally provide short-term results, in contrast to the long-term results generated by population-wide screening programs. They argue that taking the randomized trial data at face value "misrepresents the likely long-term population impact of PSA screening (relative to no screening) in the United States."

    Dr Etzioni and coauthors discuss the results of modeling studies that give a different picture of the benefits of PSA screening. Based on those models, screening may explain 45 percent of recent declines in U.S. deaths from prostate cancer, while changes in treatment account for 33 percent. When the randomized trial data are extrapolated to the U.S. population over the long term, the absolute reduction in deaths attributed to screening appears at least five times greater than in the original trial reports.

    Modeling studies also suggest a lower rate of overdiagnosis -- screening detection of slow-growing prostate cancers that otherwise would have caused no harm -- than reported in the trials. Dr Etzioni and colleagues conclude, "With a disease whose hallmark is a lengthy natural history, the harms of developing cancer screening policies based primarily on limited-duration screening trials may well outweigh the benefits."

    Counterpoint: Trials Are Best Evidence on Screening Effects But in their "Counterpoint" essay, by Dr Melnikow and colleagues notes that the U.S. and European trials provided 11 to 13 years' follow-up in more than 250,000 individuals. They also point out that the U.S. trial was highly representative of the population and showed no reduction in death resulting from annual PSA testing. (Dr Melnikow and colleagues were members of the U.S. Preventive Services Task Force when the recommendation was made.)

    They add that, because of "competing causes of death," it becomes even less likely that a large reduction in deaths from prostate cancer will appear over long-term follow-up. The chances of overdiagnosis and potential harms from screening are also likely to increase with continued aging. Dr Melnikow and coauthors conclude, "Projections from models are subject to mistaken assumptions and investigator biases, and should not be accorded the same weight as evidence from randomized controlled trials."

    In an editorial response, Dr Etzioni's group points out that modeling plays an essential role in addressing questions about the harms and benefits of screening. "While we acknowledge the centrality of screening trials in the policy process," they write, "we maintain that modeling constitutes a powerful tool for screening trial interpretation and screening policy development."

    The debate is "no mere academic exercise," according to an editorial by Ronnie D. Horner, PhD, of University of Cincinnati Medical Center. With the increased emphasis on disease prevention under health care reform, it is essential to offer those services most likely to represent value -- including cancer screenings. While there's no easy answer, Dr Horner writes, "I am hopeful that this Point-Counterpoint exchange will initiate a discussion among healthcare scientists that will yield greater guidance for determining whether a health care service is, indeed, value health care."

    A group representing families of those killed at the Columbine, Virginia Tech and Aurora mass shootings wants to ensure any unspecified funds raised as a result of the Newtown shooting go directly to victims and their families.

    Newtown, the Connecticut town where a gunman killed 20 children and six adults at Sandy Hook Elementary School on December 14, has identified more than 60 funds raising money on behalf of victims or projects related to the shooting.

    The families of some mass shooting victims want a National Compassion Fund established to manage future donations.

    "Going back to Oklahoma City, we've seen families who have had to endure not only horrific loss, but also the unimaginable task of wrestling with Byzantine nonprofit bureaucracies to access financial relief intended for them," the families said in a statement. "It's time to stop the madness. We cannot watch this happen, yet again, in Sandy Hook."

    The informal group, so far unnamed, has initiated talks with senior White House officials and two members of Congress about establishing an official fundraising operation for such tragedies, said Caryn Kaufman, a spokeswoman for the families. She declined to name the officials out of concern it might jeopardize the project.

    The group is examining whether legislation or an executive order would be needed to create a federal or nonprofit entity that would coordinate donations after any future tragedy.

    The project has won initial support from Kenneth Feinberg, the influential Washington lawyer who administered funds for victims of the September 11, 2001, attacks and several mass shootings.

    Jerri Jackson, whose son Matt McQuinn was killed with 11 others in the 2012 movie theater shooting in Aurora, Colorado, wants it to be easier for future victims to receive aid.

    "It's not a matter of if something else happens, it's when something else happens," Jackson said. "Immediately, a fund would be set up. It would be a trusted fund that people would feel they could give to and the money would go to the victims."

    Jackson said she was forced to "jump through hoops in the midst of tragedy" to receive money raised in her son's name.

    Nearly $5.9 million in Aurora donations were channeled to the Community First Foundation. When victims' families publicly complained about the pace of disbursement, Colorado Governor John Hickenlooper called in Feinberg, who oversaw the disbursement within 45 days. The victims' fund was later shut down.

    After the 2007 Virginia Tech University shooting that killed 32 people, more than $10 million spontaneously flowed to the university's fundraising foundation, university spokesman Larry Hincker said.

    Because that foundation was only authorized to spend money on the university, an executive order from the governor was needed to disburse the funds to victims, Hincker said.

    Michael Pohle, a member of the group whose son was killed at Virginia Tech, said: "What was so insulting was we had to fill out documents, have them notarized, and basically beg and apply for dollars."

    Feinberg recognized some grieving survivors were upset but defended the disbursement process following both tragedies, noting the highly unusual circumstances, complex legal requirements and emotional strain on the families.

    FAMILIES BAND TOGETHER

    The ad hoc group of 64 people lost relatives in tragedies such as the September 11 attacks, the 2008 shooting at Northern Illinois University and the Sikh temple shooting last year at Oak Creek, Wisconsin.

    The group has not involved the Sandy Hook families regarding the distribution of Newtown funds, saying it was too soon for those still grieving. Even so, the group was concerned about the confusion caused by having dozens of nonprofit groups collecting money in Newtown, said Kaufman, the group's spokeswoman.

    Some of the Newtown funds were designated for a specific purpose, such as creating a playground or scholarship. In other cases, the funds refer to more general goals of supporting the victims. The group believes that money should go to the victims' families.

    Feinberg has overseen disbursements of funds following September 11, the BP oil spill and several mass shootings. Though he is not affiliated with the group, he supports the concept Of having a protocol in place when a future tragedy occurs.

    "I think it's a great idea," Feinberg said. "The question is: Is there the political will to do it?"

    Tax Deductions: Hidden Gems and Hazards

    Congressmen and pundits who have lately singled it out as a target in national debt talks know that it's a politically perilous road.

    For now, the deduction's there, so make the most of it. In addition to payments for mortgage interest and taxes, premiums on mortgage insurance and interest charges on late payments are deductible, advises Smith.

    A little-known break for home buyers, Smith says, is that even if your seller paid the "points" for your home mortgage as part of the closing, that prepaid interest is still deductible for the buyer.

    Also, at a time when many homeowners are refinancing thanks to low interest rates, keep in mind that you must amortize the points over the life of the loan, explains Mary Canning, dean of the Schools of Taxation and Accounting at Golden Gate University.

    That is, if in your second year of owning a home you refinance and pay one point on a $500,000, 30-year loan, you're limited to amortizing the $5,000 of points over 30 years.

    The amortization begins in the month the refinancing is done and points are paid. So if the refi occurs in September the calculation is this: Four months in the year from September, divided by 360 months of the loan, multiplied by 5,000 in points (4/360 x 5000)?which equals $56 in eligible deductible points.

    If in year 3 you refinance again in January, you've retired that earlier loan and can deduct the unamortized points of $4944 ($5,000 minus $56).

    Charity: Good Will Gives Back

    Most people don't forget to deduct their charitable contributions, says Canning. "But many miss the mileage," she says. Driving you've done as volunteer?whether for Meals on Wheels or picking up an elderly neighbor for church?can be deducted at 14 cents a mile. (You cannot deduct the cost of driving yourself and your family to church?that's not an act of charity.)

    It's best if you've kept a mileage log. But in a pinch, if you have a record showing you made the trips and Google Maps to calculate the distance, it's still a legitimate deduction.

    In the same vein, a scout leader can deduct the cost of her uniform and a volunteer can deduct the miles for visiting a shut-in.

    More good deeds that reap tax benefits: Deduct expenses?mileage and more?for a volunteer excursion for Habitat for Humanity or the like, given you didn't do any vacationing along the way. And housing a foreign exchange student is worth up to $50 a day.

    A churchgoer can deduct the amount of his tithe?but only if he's got a record of it. And for any amount over $250, that means a receipt stipulating that the giver received no benefit for his contribution. Even under the threshold, use checks so you have a record. Unsubstantiated claims of cash in the collection plate don't fly.

    Red flags: Claims of large gifts of appreciated property get the attention of the IRS. Also, if you donate a box of clothing to the Salvation Army, the IRS assumes everything included is valued for a thrift shop. So you may have paid $1,000 for a dress, but that doesn't mean you can claim a $500 value when you donate it. The only way around this is a professional appraisal.

    Work: On-the-Job Deductions

    Many Americans have been on the job hunt, and the government allows deductions for that pursuit, but only if those expenses?which include resume preparation services, printing and mailing costs, travel expenses, etc.?exceed 2 percent of gross income. And if it's your first job hunt, you don't qualify.

    If you landed the job but had to relocate, you can deduct unreimbursed moving expenses. Keep in mind that the new job must be 50 miles farther from your previous residence than your old office.

    For the self-employed, there's a lot to track. A home office can make for a handsome deduction, but auditors have been known to demand proof of the space and its exclusive business use. Keep records and don't get cute.

    However, Canning says deductions for a legitimate home office can go beyond the obvious; don't forget bills for water, garbage, housekeeping and even gardening, with all amounts determined by the percentage of the residence taken by the workspace.

    Also, consider business-related expenses of your automobile, depreciating office equipment, continuing education, and entertainment and restaurant tabs.

    For cellphones and computers, your best bet to avoid the auditor's red pen is by having ones you use exclusively for work. Mixed-use devices muddy the waters.

    Holding open houses may also be included in your Texas Realtor?s marketing plan. But if not, or if you would prefer not to, you can always discuss these options with them and they will be happy to accommodate your wishes. These are great conversations to have with your Texas Realtor, whose goal is to help you get the most out of your investment.

    Asking for referrals is a great way to search for a Texas Realtor. Ask your friends, family members or coworkers who they would recommend. A word-of-mouth referral is great starting point where you can get honest feedback.

    How to See Argo Tonight (the Constellation, Not the Movie)

    Spring has just arrived in the Northern Hemisphere, bringing with it some new and interesting skywatching sights.

    The evening sky of early spring is dominated in the northeast by a very large constellation, Ursa Major, the Great Bear, and in the southeast by Leo, the Lion, containing the bluish-white 1st-magnitude star Regulus.?

    Ursa Major is indeed one of the largest constellations, but at one time there was a constellation in our spring sky that was even bigger. It's the star pattern that the ancients made into the huge constellation of Argo Navis, the Ship, which currently occupies most of the sky down near the south and southwest horizon in early evening.

    Was this Noah's Ark?

    If you mention the name "Argo" days, the first thing that will probably pop into most people's heads is the Oscar-winner for best motion picture. [Sky Maps for March 2013]

    Indeed, you'll be looking in vain for the name Argo on modern sky charts, for it officially passed out of existence in 1930, when the International Astronomical Union defined the presently recognized constellations and their boundaries.?

    The IAU broke Argo up into four separate parts: Puppis the Stern, Vela the Sails, Carina the Keel (which contains the brilliant yellow-white star Canopus, the second brightest in the night sky), and Pyxis the Mariner?s Compass. The stars that comprise this last group actually belonged at one time to the non-defunct star pattern Malus, the Mast.

    Interestingly, in his classic star guide "The Stars ? A New Way to See Them" (Houghton-Mifflin Co., Boston), the late H.A. Rey kept Argo intact, referring to it simply as "The Ship," and describing it as ?a large majestic group.?

    Argo was regarded as a ship by several early civilizations and has often been thought of as Noah's Ark. About 5,000 years ago it appeared farther north than it does now, thanks to the wobbling motion of the Earth on its axis known as precession. ?

    Ancient stargazers in Greece and Asia Minor would see this celestial ship sailing westward ? albeit stern first (Rey suggested that perhaps the ship was trying to correct an error in navigation) ? and along the Mediterranean horizon as the night progressed.?

    In Greek mythology, the Argo carried Jason and his 50 Argonauts to Colchis at the eastern end of the Black Sea, to recover the Golden Fleece. The goddess Athena, who procured a magical oaken prow that had the powers of thought and speech and was thereby able to guide the crew, supervised the ship?s construction.?

    At launching time, however, the vessel refused to slide into the water until Orpheus charmed it with his lyre, one of a number of feats he performed with that instrument (now commemorated in our summer sky by the constellation Lyra).

    Don?t follow this cross!

    Readers of this column who live south of the equator are probably well acquainted with four stars in this part of the sky ? two belonging to Vela and two belonging to Carina ? known collectively as the "False Cross."?

    Not only do these four stars bear a superficial resemblance to the true Southern Cross (Crux), but they are even oriented in roughly the same way, though positioned farther to the north.

    Hence an uninitiated observer might easily mistake the False Cross for the true one, which is honored on the flags of such countries as Australia and New Zealand. But only Crux points to the south celestial pole (matching the Pointers of the Big Dipper in this respect).

    Yet the False Cross will have its time of glory 66 centuries from now, in the year 8600, when the pole will have precessed to a point just east of the cross-arm' intersection.? By then, perhaps when our descendants are asked where the South Pole of the sky is located, all they need do is say, "X marks the spot!"

    100 tea bags a day causes rare bone disease in woman

    A 47-year-old Michigan woman developed a bone disease rarely seen in the U.S. after she drank a pitcher of tea made from at least 100 tea bags daily, for 17 years, researchers report.

    The Detroit woman visited the doctor after experiencing pain in her lower back, arms, legs and hips for five years.

    X-rays revealed areas of very dense bone on the spinal vertebrae and calcifications of ligaments in her arm, said study researcher Dr. Sudhaker D. Rao, a physician at Henry Ford Hospital who specializes in endocrinology and bone and mineral metabolism.

    The researchers suspected the woman had skeletal fluorosis, a bone disease caused by consuming too much fluoride (a mineral found in tea as well as drinking water).

    The patient's blood levels of fluoride were four times higher than what would be considered normal, the researchers said.

    Skeletal fluorosis is endemic in regions of the world with naturally high levels of fluoride in drinking water, including some parts of India and China, but is rare in Europe and North America. (Low levels of fluoride are added to drinking water in the United States to prevent cavities, but aren't high enough to cause fluorosis.)

    Rao said the patient was originally referred to him because her doctors suspected she had cancer, which can also show up on an X-ray as areas of dense bone. But because Rao had seen cases of skeletal fluorosis in his native India, "I was able to recognize it immediately," he said.

    Excess fluoride is typically eliminated from the body by the kidneys, Rao said. But if one consumes a lot of it, as this patient did through tea drinking, over time, the fluoride forms crystal deposits on bone, Rao said.

    A few other cases of skeletal fluorosis caused by tea drinking have been reported in the United States. In these cases, patients typically drank a gallon of tea a day, Rao said. Rao and colleagues instructed their patient to stop drinking tea, after which she experienced an improvement in symptoms. The fluoride deposits will gradually go away as the bone remodels (or repairs) itself, a process that occurs frequently in the body, Rao said.

    A description of the case will published tomorrow (March 21) in the New England Journal of Medicine.

    ? New Chinese Premier Li Keqiang dismissed hacking accusations against China as "groundless" on Sunday and said his government was committed to strong ties with Washington.

    Referring to allegations that China's military was behind massive hacking attacks on U.S. entities, Li reiterated Beijing's statements that China is a major target of global hackers and opposes all such criminal activity.

    "I think we should not make groundless accusations against each other but spend more time doing practical things that will contribute to cybersecurity," Li said in his first news conference in his new role.

    Li is the highest-ranking official to comment on the hacking claims made by U.S. cybersecurity firm Mandiant that provided a detailed picture of alleged cyberspying carried out by a People's Liberation Army unit based in Shanghai.

    Li said that despite their differences, conflict between the world's largest and second-largest economies is not inevitable as long as the countries respect each other's major concerns and manage their differences.

    China's new leaders "attach great importance" to relations that meet the "fundamental interests of people in both countries and serves the global trend of peace and development," Li told reporters at the traditional premier's news conference that follows the close of the annual legislative session.

    China-U.S. ties have weathered a series of crises over the past year over dissidents, Chinese trade practices, opposition to Chinese investment in the U.S. and - most recently - hacking accusations. While basically stable, political ties are seen as lagging behind the economic relationship, with two-way trade hitting almost $500 billion last year, and China's new president, Xi Jinping, isn't expected to meet President Barack Obama until an economic summit in Russia in September.

    Following Xi's elevation, Li was appointed premier last week, with running China's economy his main responsibility.

    A federal court of appeals handed a victory -- although a very limited one -- to the ACLU on Friday in the civil rights group's effort to use the Freedom of Information Act to get documents from the CIA about US drone strikes overseas.

    A three-judge panel of the DC Court of Appeals ruled unanimously that the CIA cannot simply refuse to respond by saying that it cannot confirm or deny the existence of any records. That position, the court said, has been completely undercut by public statements about the drone program made by President Barack Obama, CIA Director Leon Panetta and White House counterterrorism adviser John Brennan.

    "Given these official acknowledgments that the United States has participated in drone strikes, it is neither logical nor plausible for the CIA to maintain that it would reveal anything not already in the public domain to say that the Agency 'at least has an intelligence interest' in such strikes," the court said.


    Those statements, the court said, make it "implausible that the CIA does not possess a single document on the subject of drone strikes."

    But Friday's court victory does not hand the ACLU the key to the documents.

    The court sent the case back to a federal judge to decide whether the CIA can still argue that actually handing over any documents it has would damage national security. In fact, Friday's decision even holds out the possibility that the CIA may not have to be very explicit at all in saying what documents it has and why it wants to withhold them.?

    The ACLU filed a Freedom of Information Act request with the CIA three years ago, seeking records pertaining to the use of drones by the CIA or the armed forces for targeted killings overseas. ?When the agency failed to respond in time, the ACLU went to court.? A federal judge accepted the CIA?s argument that even answering the question of whether it had any drone records would raise national security problems. ?That ruling was reversed by the appeals court.?

    Jameel Jaffer, the ACLU lawyer who handles the case, called Friday's decision an important ruling.

    ?It requires the government to retire the absurd claim that the CIA's interest in the targeted killing program is a secret, and it will make it more difficult for the government to deflect questions about the program's scope and legal basis. It also means that the CIA will have to explain what records it is withholding, and on what grounds it is withholding them," he said.

    Novel chemical that controls cell behavior: First-in-class chemical compound might control metastases

    Mar. 13, 2013 ? It's the spread of the original cancer tumor that kills most people. That's why cancer researchers vigorously search for drugs that can prevent metastases, the spread of cancer. The research team co-led by Angela Wandinger-Ness, PhD, and Larry Sklar, PhD, at the University of New Mexico Cancer Center has found a chemical compound that appears to control cell migration and adhesion, two important characteristics of metastatic cancer cells. The team recently published a paper describing how the first-in-class compound acts on various cells.

    Dr. Wandinger-Ness, a UNM Professor of Pathology and Director of the Fluorescence Microscopy and Cell Imaging Shared Resource, studies proteins called GTPases. GTPases act like chemical switches to control how cells behave: how much a cell grows, what shape it assumes, when it enters the next growth stage, and how tightly it sticks to its surroundings, among several hundred other things. Dr. Wandinger-Ness was interested in a particular GTPase called Cdc42; it controls cell migration and cell adhesion. "It's an important target in many diseases," says Dr. Wandinger-Ness. "Cancer is just one. But there were no compounds that target this GTPase." So she collaborated with Dr. Sklar and Tudor Oprea, MD, PhD, to find a compound that did. And they were successful.

    Dr. Sklar is a UNM Professor of Pathology and co-Leader of the Cancer Biology and Biotechnology Research Group at the UNM Cancer Center. He created and now oversees the UNM Center for Molecular Discovery. Dr. Oprea is a UNM Professor of Medicine and co-Director of the Flow Cytometry & High Throughput Screening Shared Resource at the UNM Cancer Center. He analyzed Cdc42 using three-dimensional molecular rendering software. The team used Dr. Oprea's analysis of Cdc42 to visualize how a compound might interact with the Cdc42 GTPase to stifle its activity. Then they searched for such a compound in the UNM Molecular Discovery library.

    The search process was akin to finding a needle in a haystack, but the latest high throughput flow cytometry equipment and molecular rendering software available at the UNM Cancer Center speeded their analysis significantly. The team analyzed thousands of compound candidates by first narrowing their search to the few hundred likely candidates and then testing those against several kinds of GPTases at a time. "From a purely discovery perspective, that's a high impact, novel way to look for small molecules," says Dr. Sklar. The compound they found is called CID2950007.

    Structurally similar to NSAIDs -- non-steroidal anti-inflammatory drugs -- CID2950007 restrains the Cdc42 GTPase from changing a cell's cytoskeleton. Much like a skeleton gives a human body shape, a cell's cytoskeleton keeps the cell from collapsing on itself. The cytoskeleton enables a cell to move by growing amoeba-like legs called filopodia. Cdc42 also helps to keep cells where they need to be by enabling them to adhere more tightly to their surroundings. So, while uncontrolled growth and movement are hallmarks of metastatic cancer cells, growth and adherence are important traits for healthy cells. Tightly controlling just how Cdc42 causes a cell to behave is crucial.

    In the paper published February 4, 2013, the team of scientists report that CID2950007 was the only compound they found that affected the Cdc42 GTPase without affecting any other GTPases. This selectivity is important to control the compound's effects on a cell. They also found that the compound works by changing the physical structure of Cdc42, so it doesn't destroy Cdc42 but it does control how Cdc42 interacts with other proteins in the cell. Their studies showed that CID2950007 decreased filopodia growth and cell adhesion in ovarian cancer cells and prevented cell adhesion in white blood cells. And their studies demonstrated that CID2950007 blocked Hantavirus infection in monkey kidney cells. By affecting the Cdc42 GTPase, and thus the cytoskeleton, CID2950007 has the potential to fight not only cancer but also infectious diseases.

    Human use of CID2950007 as a cancer drug is a long way off. Before the Food and Drug Administration approves any drug for human use, it first requires the successful results of several toxicity and dose escalation studies on several types of animals. Then, the clinical trials process, which can take over 10 years, may begin.

    "There are going to be a lot of side effects because these adhesion proteins have many other functions," says Dr. Sklar. So refining CID2950007 into a drug will take further collaboration and studies before toxicity studies and dose escalation studies can begin. To refine the compound, the UNM Cancer Center researchers will continue to collaborate with Jeffrey Aub?, PhD, Kansas University Distinguished Professor of Medicinal Chemistry, and Jennifer Golden, PhD, Assistant Director of the Specialized Chemistry Center at Kansas University. Still, control of GTPase Cdc42 offers promise as a way to control cancer metastasis. Says Dr. Wandinger-Ness, "there's a lot of enthusiasm for a compound like this -- because there weren't any. This is a first-in-class."

    N. Korea threat of nuclear attack not easily dismissed

    In a sign that North Korea's threats are wearing thin, their closest ally ? China -- voted with the U.S. for tough economic sanctions on luxury goods. North Korea responded by announcing they "will be exercising our right to preemptive nuclear attack." NBC's Andrea Mitchell reports.

    Thursday?s announcement by North Korea that it could launch a pre-emptive nuclear attack against the United States in the face of new U.N. sanctions is a predictable escalation of the isolated nation?s increasingly aggressive stance toward Washington over the past year. But experts note that Pyongyang?s recent advances in its nuclear weapons and missile programs mean that such bellicose rhetoric cannot be taken lightly.

    ANALYSIS

    The escalation of the North?s oratory began not long after the country?s 28-year-old leader, Kim Jong Un, ?took over from his late father, Kim Jong Il, on Dec. 28, 2011. It has been accompanied by two space launches ? one successful ? and a third nuclear weapons test.

    ?It is not unusual for the North to make threats against the U.S., Japan or South Korea. And on occasion -- as in the case of the 2010 artillery barrage of Yeonpyeong Island and an earlier attack on a South Korean gunboat -- it has carried out these threats. ?It has never taken any military action after threatening the United States, however.


    Some analysts have suggested that the latest round of threats is intended to show that the young Kim will continue his father?s legacy of hostility toward the U.S.

    To what end?

    North Korea has long wanted the U.S. to sit down with its negotiators to hammer out an agreement to end the Korean War, which ended in 1953 not in a peace treaty but in a truce.

    The North would like to gain concessions from the U.S. in such a negotiation, but its escalating threats and rhetoric have the opposite effect: ?The Obama administration, like preceding administrations, has steadfastly refused to negotiate with Pyongyang.

    This picture, released Tuesday by North Korea's official KCNA news agency, is said to show a rally by citizens and soldiers to support a statement by the Supreme Command of the Korean People's Army that it will scrap the armistice signed in 1953 that ended a three-year war with South Korea if the South and the United States continue with annual military drills.

    The problem is that North Korea, which has long taken a backseat in U.S. councils to the Middle East, does have military capabilities that could at the very least threaten U.S. interests in North Asia.

    According to a recent analysis, North Korea has a weapon stockpile that could threaten both Japan and South Korea and, in longer term, the United States. Some of the weapons have already been deployed, say U.S. officials, who spoke to NBC News on condition of anonymity. Moreover, the North has begun research into more advanced and dangerous weapons, possibly even thermonuclear weapons, they say.?

    At the high end of the stockpile range, U.S. officials and other researchers said North Korea may already have up to "a few dozen" nuclear weapons that could be fitted atop its vast fleet of ballistic missiles. Those missiles are limited to an intermediate range, capable of hitting targets in Japan, South Korea or elsewhere in the northern Pacific, including U.S. military bases as far south as Guam, the officials believe.

    The U.S. believes the space launch tests are part of a development plan for an intercontinental ballistic missile (ICBM) capable of reaching the continental United States with a payload of several hundred kilotons ? 10 to 20 times the size of the bombs that destroyed the Hiroshima and Nagasaki.

    U.S. officials publicly express confidence that the national missile defense system based in Alaska would be able to shoot down any incoming North Korean ICBM.

    ?I can tell you that the United States is fully capable of defending against any North Korean ballistic missile attack,? White House spokesman Jay Carney said Thursday in response to a question about the North Korean threat.

    ?He also said the U.N. sanctions will make it harder for Pyongyang to continue to make progress on its weapons and missiles.?

    ?North Korea ? will now face new barriers to developing its banned nuclear and ballistic missile programs,? he said. ?Resolution 2094 increases North Korea's isolation and demonstrates to North Korea's leaders the increasing costs they pay for defying the international community.??

    For the past several years, the U.S. also has been monitoring North Korean research into thermonuclear weapons ? hydrogen bombs and bombs known as boosted-fission weapons, in which plutonium and uranium are combined for a higher energy yield. (The problem is that if the North conducted a test and claimed that it was thermonuclear, the U.S. would have difficulty determining if the North was telling the truth. The test site at Kilchu is far enough inland that the U.S. would not have access to the particulate matter needed to make an accurate determination, experts say. )

    David Guttenfelder, AP's chief Asia photographer, was given unprecedented access on his 2011 journey to Pyongyang and areas outside the nation's showcase capital.

    David Albright, president of the Institute for Science and International Security, or ISIS, a nonpartisan nuclear arms research group, said last year that any tests in the future may also be about ensuring the reliability of North Korea's current weapons design.

    "Once you get beyond a dozen, it makes sense to test type and reliability of your weapons," he said. Albright said then that his group's estimate of North Korea's weapons stockpile is a bit less than those provided by the U.S. officials, but that ISIS, too, believes Pyongyang has "missile-deliverable weapons."

    The design of the weapons is believed to be based on Chinese models (as were the first generation Pakistani nuclear weapons). The design is basic, and was developed in the 1960s with help from the Soviet Union, which used it to produce a whole line of nuclear warheads.

    While some analysts suggested that the North planned its December rocket launch to gain attention ahead of the presidential election in South Korea , some in the U.S. non-proliferation community think otherwise. They expect that once the North feels comfortable with its ICBM technology, it will deploy the missiles.? They point to the Musudan intermediate range missile which was tested in middle of the last decade, then deployed ? presumably with nuclear warheads ? and aimed at Japan.

    Once the North has confidence in the long-range missile based on the space rocket, U.S. officials believe they will deploy it as well, making North Korea the third nation to have nuclear weapons targeted at the United States, after Russia and China.

    Many in the Obama administration see that as a more frightening prospect than Iran gaining nuclear weapons, believing that Tehran is a rational actor that will serve its own national interest and preserve the regime, compared to successive generations of North Korean leaders who have shown that they are unpredictable and erratic.

    But would it force the U.S. to conduct face-to-face talks with the North? State Department spokeswoman Victoria Nuland said in December that the North has a better option.

    Referring to Kim Jong Un, Nuland said: "He can plot a way forward that ends the isolation, that brings relief and a different way of life and progress to his people, or he can further isolate them with steps like this. He can spend his time and his money shooting off missiles, or he can feed his people, but he can't have both."
